Improved Dilution Plating Test for Bacterial Spot on Tomato, Pepper and Eggplant

tomatoes

Naktuinbouw Laboratories has enhanced the dilution plating test for detecting Xanthomonas pathogens responsible for ‘bacterial spot’ in tomato, pepper, and eggplant seeds.

Following dilution plating, all suspect colonies undergo confirmation using an improved real-time PCR assay. The updated markers in this assay align with those used in the seed extract PCR, enabling the detection of key taxonomic groups: Xanthomonas hortorum pv. gardneri, Xanthomonas vesicatoria, and Xanthomonas euvesicatoria pv. euvesicatoria and Xanthomonas euvesicatoria pv. perforans (the latter two remain indistinguishable).

The method involves collecting bacterial colonies from the dilution plating plates and extracting their DNA, according to a press release. Pathogen-specific markers are then used to confirm the presence a specific bacteria. This makes it possible to confirm with more certainty whether the seed samples actually contain the live pathogen.

The species Xanthomonas euvesicatoria pv. euvesicatoria, X. euvesicatoria pv. perforans, X. hortorum pv. gardneri and X. vesicatoria (formerly known as the X. campestris pv. vesicatoria complex) result in necrotic spots on all above ground plant parts and result in significant crop losses, primarily in tomato and pepper.

Xvepg on seeds of tomato, pepper or eggplant:

  • Test code: XCVA100 or XCVA300
  • Sample size: 10.000 or 30.000 seeds
  • Test fee*: € 231 resp. € 307
  • Duration test: 17 days
